چکیده مقاله:

Background and Objective: The rate of survival in acute promyelocytic leukemia (APL) can dramatically improve, if the patients receive all-trans-retinoic acid (ATRA) treatment. However, this drugchr('39')s toxicity is a major problem in APL treatment. Previous researches have demonstrated that phyto-polyphenols such as epigallocatechin gallate (EGCG) and kaempferol cause apoptosis in hematopoietic neoplasms, but do not negatively impact healthy cells. The present study compared the differentiation effects of kaempferol and EGCG, as well as ATRA in NB4 leukemia cells during five days. Materials and Methods: Herein, kaempferol and EGCGchr('39')s differentiation-inducing activity was examined by NBT assay and Real-time PCR in leukemia NB4 cells. Results: EGCG (25 µM) and kaempferol (50 µM) induced the NB4 cellschr('39') differentiation, towards a granulocytic pattern similar to ATRA (1 μM). EGCG further suppressed PML/RARα clinical markerchr('39')s expression compared to kaempferol, it also decreased HDAC1 expression in leukemia NB4 cells. Conclusion: Based on this study, compared to kaempferol, EGCG at low concentrations is preferred for long-term ATRA therapy in APL patients.

Pro-oxidative effects of tea and polyphenols, epigallocatechin-3-gallate and epigallocatechin, on G6PD-deficient erythrocytes in vitro.

Glucose-6-phosphate dehydrogenase (G6PD)-deficient subjects are vulnerable to chemical-induced hemolysis if exposed to oxidative agents. Recent studies reported that green tea and its constituents might act as pro-oxidants. Our objective was to investigate effects of tea and its polyphenolic components on the oxidative status of human G6PD-deficient erythrocytes. Erythrocytes of G6PD-deficient ...

Anti-proliferative activity of epigallocatechin-3-gallate and silibinin on soft tissue sarcoma cells

Disseminated soft tissue sarcomas (STS) present a therapeutic dilemma. The first-line cytostatic doxorubicin demonstrates a response rate of 30% and is not suitable for elderly patients with underlying cardiac disease, due to its cardiotoxicity. Well‑tolerated alternative treatment options, particularly in palliative situations, are rare. Therefore, the present study assessed the anti‑prolifera...

Prooxidant property of green tea polyphenols epicatechin and epigallocatechin-3-gallate: implications for anticancer properties.

It is believed that anticancer and apoptosis inducing properties of green tea are mediated by it's polyphenolic constituents particularly catechins. A number of reports have shown that green tea polyphenol (-)-epigallocatechin-3-gallate (EGCG) is among the most effective chemopreventive and apoptosis-inducing agents present in the beverage. Plant polyphenols are naturally occurring antioxidants...

Anticancer activities of epigallocatechin-3-gallate against cholangiocarcinoma cells

PURPOSE Epigallocatechin-3-gallate (EGCG) is an antioxidant agent derived from green tea. Because it has chemopreventive and anti-invasive effect against various cancer cells, EGCG can be used to inhibit proliferation and invasion of cholangiocarcinoma (CCA) cells. METHODS The anticancer effects of EGCG were studied using human CCA cells (HuCC-T1). Apoptosis was analyzed by Western blotting. ...

Anti-Fatigue Effect of Green Tea Polyphenols (-)-Epigallocatechin-3-Gallate (EGCG)

BACKGROUND (-)-Epigallocatechin-3-gallate (EGCG) is the most abundant of the green tea polyphenols that exhibit a variety of bioactivities. The objective of this study was to evaluate the anti-fatigue effect of EGCG by forced swimming exercise. MATERIALS AND METHODS The mice were divided into one control group and three EGCG-treated groups. The control group was administered with distilled wa...

Effect of genistein on the bioavailability and intestinal cancer chemopreventive activity of (-)-epigallocatechin-3-gallate.

The green tea (Camellia sinensis) catechin, (-)-epigallocatechin-3-gallate (EGCG), has shown cancer-preventive activity in animal models. Previously, we have reported the bioavailability of EGCG in rats and mice. Here, we report that cotreatment of HT-29 human colon cancer cells with genistein (from soy) increased cytosolic EGCG by 2- to 5-fold compared with treatment with EGCG only. Inclusion ...
